Chapter 101 Prion Research Group Formally Established(1/2)

Due to the particularity of prions, Zhou Wen was extremely careful when doing experiments.

Biomasks, biological gloves, biochemical protective clothing, fully armed.

Qin Haitao and the other four were just helping to get things done, and did not directly contact the vessel containing the virus.

There are currently four methods to detect prions.

1. Electron microscopy method.

2. Capillary SDS-gel electrophoresis.

3. Capillary electrophoretic immunoassay of fluorescently labeled peptide chains.

4. Two-color strong fluorescence target scanning method.

Among them, the second "gel electrophoresis method" is the best and most direct detection method.

It is mainly purified by physical and chemical methods-differential, ultracentrifugation, sodium n-lauroylsarcosine (lauroylsarcosine), pk treatment, and purified prpsc.

Capillary SDS-gel electrophoresis is then performed on a "capillary electrophoresis instrument".

According to the time and peak shape of the absorption peak at 214 nm wavelength when each component passes through the detector, the prion disease cause prpsc was identified.

This method uses a small amount of specimens and does not require immunologic reagents, so the judgment is very intuitive.

The disadvantage is that it requires two devices such as "ultracentrifuge" and "special capillary electrophoresis instrument".

Ultracentrifuges are currently basically purchased foreign brands, with prices starting from 500,000 rmb.

Prion-specific capillary electrophoresis instruments are also very expensive, requiring more than 200,000 rmb.

It’s not that the price is expensive, but the key is that there are few opportunities to use it.

Generally, hospitals cannot even make money from purchasing equipment, and only rich Grade A hospitals like Jiangzhou Hospital will purchase.

Because of this, medical resources in big cities are scarce.

A small city has a strange disease, and you can't even get a test.

Zhou Wen carefully prepared reagents step by step according to physical and chemical methods, extracted the diluent, and placed them on an ultracentrifuge to separate the mutated prion protein.

The newly upgraded biological immunology made him more comfortable.

During this process, Zhou Wen also taught Qin Haitao and Dong Wenying, telling them what to pay attention to during the physical and chemical test of routine prions.

"When adding lauroyl, you must remember..."

Not to mention university professors, even experts from domestic prion institutes cannot explain these experiences clearly.

Because China is basically in its infancy in viruses, especially prions research, these practical experiences can only be mastered by top foreign research institutes.

For example, the Institute of Yeast Prions at the University of Kent, UK.

For a long time, the new discoveries in the field of prion protein have been "monopolized" by several research teams in the field of yeast.

Those top research institutes will not easily teach you technology.

Unless you can get their offer.

Qin Haitao and the others obviously did not realize how lucky they were. They just felt that Zhou Wen’s explanation was very easy to understand, and even told them the key points of the action.

When Zhou Wen said that, Qin Haitao and Dong Wenying listened attentively.

As for the other two old experimenters, although they were listening on the surface, they actually didn't listen at all.

Because they felt that it was impossible to extract prions with Zhou Wen's method, such teaching was simply a talk on paper.

Zhou Wen did not have an "electric eye", how could he know these specific experimental variables? Everything was just his guess.

After purifying the prpsc, Zhou Wen injected the prion-containing diluent on Beckman P/ace 5 500 and conducted an electrophoresis experiment.

"Okay~" After the injection, Zhou Wen breathed a sigh of relief.

"Qin Haitao, please stay and take care of the machine, and everyone else washes their hands and eats."

When everyone heard this, they went to the sink to wash their hands...

After dinner, Zhou Wen didn't dare to go anywhere and went back to the laboratory.

The samples obtained by conventional extraction methods are too rare, and I feel really worried if I don’t watch them in person…

At 7:30 pm, the electrophoresis experiment ended.

One of the experimenters, Huang Xingzhong, took the electrophoretic analysis sheet and glanced casually, subconsciously thinking that this experiment would fail as before.

However, the next second, a look of surprise appeared on his face, "Hey, come and see, it seems that the prions have been detected..."

When the rest of the people heard this, they surrounded them.

"Hey, it's really awesome."

"Teacher, come and see, we have detected the prions."

"Shit... it shouldn't be, how could this be possible? Did you make a mistake..."

Zhou Wen took the analysis order and looked carefully, and soon he could not suppress his joy on his face.

No matter how smooth the previous experiment was, nothing was 100% before the results came out.

For example, if a failure of the capillary electrophoresis instrument will also lead to detection failure.

This will make all previous efforts in vain.

So sometimes, luck also accounts for part of the factor when doing experiments.

After confirming that it was not a dazzle, everyone's faces showed an indescribable look of excitement.

To be honest, no one, including Qin Haitao and Dong Wenying, think that prion protein can be extracted under conventional methods.

It is really a very proud thing to extract prion proteins without expectation.

After the excitement, Qin Haitao immediately notified Yang Liqun and Zhang Shuguang.

After hearing this, the two rushed over immediately.

"According to the test, this is an aggregate prion..."

Yang Liqun and Zhang Shuguang looked at Ming's clear electrophoresis analysis sheet, their faces were filled with surprise, joy, excitement, excitement and incredible expressions.

"Is this really detected in urine?"

"It's the Zhang Group! The sample was collected by Zhou Wen and I."

Hearing Dong Wenying's words, Yang Liqun and Zhang Shuguang looked incredible.

In the case of routine and unconventional testing, the extraction of prions has completely different meanings.

This is a very important discovery and has high research value.

"You are all good. Although this is just a small step in prions research, it is a big step for our China in the field of prions."

"Thank you for the praise of Team Yang. It's mainly because of Teacher Zhou. If he hadn't been persisting, we might have given up just by seeing it."

Yang Liqun and Zhang Shuguang looked at Zhou Wen.

Both of them have always been very optimistic about this young man, and Zhou Wen has not disappointed him. He has shown a very strong talent in biomedical science.

And they often bring some surprises.

For example this time.

Yesterday they predicted that it is absolutely impossible to extract prion proteins under conventional collection methods.

As a result, Zhou Wen was slapped in the face the next day.

But this face feels comfortable.

If possible, they hope he will continue to slap in the face, and it is better to shoot the left and right!

After being excited, Yang Liqun and Zhang Shuguang went to the computer to analyze and study the sample collection records, hoping to find a certain rule from it.

Zhou Wen also admires Yang Liqun and Zhang Shuguang's behavior very much.

This is the spirit that scientific research should have.

Although no one wants to collect prion proteins in a conventional state, they cannot be ignored because they do not want them to be studied.

Only by knowing which factors will cause prion proteins to be excreted through body fluids can we better prevent and control them.

The next morning, Yang Liqun reported Zhou Wen's discovery to the hospital and requested the formal establishment of a "prion research group" under the testing center.

The hospital leaders held a special meeting to discuss whether to approve the establishment of this research group?

As the First People's Hospital affiliated to Jiangzhou University, it is actually not too bad in terms of hardware equipment or talent reserves, but why can't it compare to other hospitals?

The main reason is that Jiangzhou First Hospital is well-versed and loose, and there is no "unique skill" that can be used to show off.

For example, the Jinling Military General Hospital in the province is well-known in terms of kidney disease.

For example, Huashan Hospital in Zhonghai is very awesome in the Dermatology, Infectious Diseases and Neurology Departments.

For example, Kyoto University First Hospital is invincible in the Department of Urology.

There is also the orthopedic department of Long March Hospital...
