Chapter 21 Experimental Procedures
With the final step of synthesizing endorphins completed, 10ml of endorphins were freshly synthesized in the test tube in Zheng Li's hand.
There are four types of endorphins - α, β, γ, and δ. The activity of β-endorphin is 5-10 times that of morphine, and the activity of the other three endorphins is the same as that of morphine.
The first discovery of endorphins by humans dates back to 1975. This substance is secreted by the hypothalamus of vertebrates in conjunction with the pituitary gland.
It was not until 2019 that Zheng Li discovered the path to artificially synthesize endorphins. What Zheng Li synthesized was β-endorphin, which is the most active endorphin.
And in the paper, Zheng Li demonstrated through experiments that β-endorphin can have an analgesic effect through external administration.
Since endorphins are automatically synthesized by the pituitary gland, people previously only knew that they had analgesic effects, but it was not known whether their analgesic effects could be effective when injected outside the body.
Trivia: Humans discovered the artificial synthesis method of morphine in the 1810s, when Zeltina discovered that morphine crystals could be separated by soaking opium in hot water and ammonia.
The molecular formula of morphine was deduced in 1847, its structural formula was determined in 1925, and its absolute configuration was proved in 1968.
And until two hundred years later, humans still have not been able to study the synthesis route of endorphins with similar effects, let alone the structural formula and absolute configuration.
It is precisely because of the difficulty of this work that Zheng Li was able to publish a paper in nature as a pure newcomer.
The reason why it is so difficult is simple. The molecular formula of morphine is O3 and its molecular weight is only 285.3, while the molecular weight of α-endorphin is 1745.96 and the molecular weight of β-endorphin is 3400.
The molecular weight of β-endorphin is more than ten times that of morphine, but the difficulty of synthesis does not increase linearly, it increases exponentially.
After Zheng Li published his paper on the artificial synthesis method of β-endorphin, Li Xulin, inspired by the synthetic route of β-endorphin, successfully developed the artificial synthesis method of α-endorphin and successfully published the paper on Cell Sub.
published.
Just relying on the synthesis methods of several other endorphins is enough to publish many articles in cell sub-journals and even cell main journals. This can be regarded as the legacy left by Zheng Li to the School of Biology of Jiangcheng University.
Zheng Li put the newly synthesized endorphins into an ultra-low temperature refrigerator, and then continued to synthesize another polymer compound.
Zheng Li used double antibody immunoprecipitation of α-endorphin and adrenocorticotropic hormone (ACTH) antiserum to inject it into pituitary tumor cells of mice.
Ordinary white mice can be bought for a few yuan, while specially bred cancer-stricken mice cost three to four hundred yuan, with designated cancer cell survival areas. For example, the mice with tumor cells in the pituitary gland required by Zheng Li cost more than one thousand yuan.
Zheng Li continued to put the culture together with the 3H-labeled amino acids, and aliquoted samples of the immunoprecipitation medium.
Sodium dodecyl sulfate/polyacrylamide gel electrophoresis of [3H]phenylalanine-labeled immunoprecipitates prepared with endorphin antiserum separated three forms with apparent molecular weights of 31000, 11700 and 3500
Endorphins; immunoprecipitates prepared with ACTH antiserum contained four forms of ACTH with apparent molecular weights of 31,000, 23,000, 13,000, and 4500.
Sequential immunoprecipitation of culture media with ACTH antiserum and endorphin antiserum (or vice versa) showed that both antisera precipitated the same 31,000 protein macromolecules.
Purified pools of different forms of ACTH and endorphins were prepared by immunoprecipitation and gel filtration.
Finally, the lipotropic tryptic peptide [βLPH(61-69)] was produced, which contains the opioid-active methionine-enkephalin sequence.
Zheng Li looked at the freshly baked trypsin peptide and showed a slightly bored smile. This kind of experiment was too childish for him.
Endorphins are a very magical compound hormone, and humans' understanding of them is still very superficial.
The biological achievements of the mage civilization far exceed the imagination of Blue Star humans, and they can conduct human experiments by cloning humans.
In the Master's logic, humans transform nature rather than nature transforming humans. In Zheng Li's view, the environmental issues discussed by Blue Star in recent years are purely false propositions.
The inability of plastic to decompose, climate warming, the spread of viruses, and the lack of fossil fuels can all be solved through technical means, rather than giving up food due to choking.
Zheng Li couldn't understand some of the strange phenomena of Blue Star. The stone tools, pottery, copperware, and ironware discarded after human use have been buried in the ground for hundreds of years. They have not decomposed and degraded for thousands or even tens of thousands of years. In modern times, cement has been widely used.
After being discarded, their creations will not degrade naturally unless they are knocked or damaged, but they want plastic to degrade in just a few years, or even without using plastic.
Having said that, endorphins and lipoprotein tryptic peptide [βLPH (61-69)] are injected into the human body together to reduce human addiction and tolerance to drugs.
You must know that after long-term use of many drugs, humans will become dependent on drugs, which is drug addiction, such as stimulants, analgesics, hallucinogens, etc.
Scientists have previously tried to reduce addiction through various means, such as cognitive behavioral therapy, oral topiramate treatment, oral dextroamphetamine sustained-release agents, and other methods.
But none of these methods have very good results.
This has a broader market than endorphins, and Zheng Li plans to wait until later to take it out.
...
"You are here. I will take you to meet the current general manager of the company. After that, I will stay in the laboratory downstairs for a long time. You will report to me about the company's operating dynamics every week."
"Usually you are responsible for listening to Cheng Gang's arrangements. Cheng Gang is our chairman. What you need to do is to study and become familiar with the company's operating mode as soon as possible."
After Zheng Li took off his gloves and white coat, he came to his exclusive office upstairs and saw Li Miaomiao who had just landed and rushed over.
Li Miaomiao, who was originally worried, calmed down after meeting Zheng Li, especially when he found that Zheng Lihe had not changed at all in school and was still immersed in research.
She nodded and said: "Okay."
Zheng Li: "Cheng Gang, this is Li Miaomiao, my classmate at Jiangcheng University. She is mainly here to serve as my secretary. You usually call her whenever you have anything to do and familiarize her with the company's operations."
Cheng Gang looked at the young girl in front of him with good looks. He thought to himself: Could it be that Director Zheng wanted to have an office romance and brought his girlfriend to the company?
If this is the case, Cheng Gang will lower his evaluation of Zheng Li by at least two levels.
However, he remained calm on the surface: "Okay, I will make arrangements for Ms. Li. I am Cheng Gang. We will have a pleasant cooperation from now on."
Cheng Gang stretched out his hand to shake hands with the other party. When Li Miaomiao shook hands, he said: "I would like to thank Mr. Cheng in advance for taking care of him. I hope we can have a happy cooperation in the future."
Zheng Li continued: "Cheng Gang, I understand the doubts in your heart. Let me make it clear. Li Miaomiao and I are just classmates. To be precise, we are very good friends, but we are not boyfriend and girlfriend."
"The reason why I asked her to help me is because the company's management is all people sent by Mr. Lei. I need someone I can trust to prevent Mr. Lei and I from falling out, which will not affect the normal operation of the company.
