Chapter 21 Experimental Procedures
With the completion of the last step of synthesizing endorphins, Zheng Li's test tube was freshly synthesized 10ml of endorphins.
There are four types of endorphins - α, β, γ, and δ. Among them, the activity of β-endorphins is 5-10 times that of morphine, and the activity of the other three endorphins is consistent with that of morphine.
The first discovery of endorphins in humans dates back to 1975. The hypothalamus of vertebrates works together with the pituitary gland to secrete.
It was not until 2019 that Zheng Li discovered the path to artificially synthesize endorphins, among which Zheng Li synthesized beta-endorphins, which is the most active endorphins.
In addition, in the paper, Zheng Li demonstrated through experiments that β-endorphins can have analgesic effects through external treatment.
Since endorphins are automatically synthesized by the pituitary gland, people previously only knew that they had analgesic effects, but they did not know whether their analgesic effects could work in in vitro injection.
Cold Knowledge: Humans discovered that morphine was artificially synthesized in the 1710s, when Zertina discovered that opium could be soaked in hot water and ammonia to separate morphine crystals.
The molecular formula of morphine was derived in 1847, its structural formula was determined in 1925, and its absolute configuration was proven in 1968.
Until two hundred years later, humans still failed to study the synthetic path of endorphins with similar effects, let alone structural formulas and absolute configurations.
It is precisely because of the difficulty of this work that Zheng Li was able to publish a paper on nature as a pure newcomer.
The reason why it is so difficult is very simple. The molecular formula of morphine is c17h19no3, with a molecular weight of only 285.3, while the molecular weight of α-endorphin is 1745.96, and the molecular weight of β-endorphin is 3400.
The molecular weight of β-endorphin is more than ten times that of morphine, but the difficulty of synthesis does not increase linearly, it increases exponentially.
After Zheng Li published his paper on artificial synthesis method of β-endorphins, Li Xulin successfully studied the artificial synthesis method of α-endorphins through the synthesis path of β-endorphins, and successfully published the paper in the cell sub-job.
Just relying on the synthesis of several other endorphins, it is enough to publish many cell sub-issues and even cell main issues. This is also the legacy left by Zheng Li to the School of Biology of Jiangcheng University.
Zheng Li put the newly synthesized endorphins into an ultra-low temperature refrigerator and then continued to synthesize another polymer compound.
Zheng Li used α-endorphin and corticotropin (actth) antisera dual antibody immunoprecipitation into pituitary tumor cells of mice.
Ordinary mice can buy one for a few dollars. Specially cultivated cancer-related mice cost 300 to 400 to specify the survival area for cancer cells. For example, the mice with tumor cells in pituitary gland required by Zheng Li to have more than 1,000.
Zheng Li continued to put the culture together with the labeled amino acids for 3h and aliquots of the immunoprecipitation medium.
Three forms of endorphin with apparent molecular weights of 31000, 11700 and 3500 were separated by endorphin antiserum by endorphin antiserum; the immunoprecipitates prepared with antiserum contain four forms of acth, with apparent molecular weights of 31000, 23000, 13,000 and 4500 respectively.
Sequential immunoprecipitation of the culture medium with sequential immunoprecipitation with both acth and endorphin antiserum (or inverse order) showed that both antiserum precipitated the same 31,000 protein macromolecules.
Purification pools of different forms of actth and endorphins were prepared by immunoprecipitation and gel filtration.
Finally, a lipoprotein trypsin peptide [βlph(61-69)] was produced, which contains the opioid active methionine-enerphalin sequence.
Zheng Li looked at the freshly baked trypsin peptide and smiled a little bored. This kind of experiment was too pediatric for him.
Endorphins are a very magical compound hormone, and humans still have a very simple understanding of them.
The mage civilization's biology is far beyond the imagination of Blue Star humans. They can conduct human experiments by cloning people.
In the logic of the mage, humans transform nature rather than nature transform humans. In Zheng Li's view, the environmental protection issues that Lan Xing has discussed for years are purely false propositions.
Plastics cannot be decomposed, climate warming, virus flooding, and lack of fossil fuels. These problems can be solved through technical means, rather than being slaughtered.
Zheng Li couldn't understand some of the strange phenomena of Lanxing. The stone, pottery, copper and iron that humans discarded after use were buried in the ground for hundreds of years, and have not been separated or degraded for thousands or even tens of thousands of years. In modern times, the creations that used a large amount of cement in modern times will not degrade naturally if they are not destroyed or damaged after being damaged. However, they want plastics to degrade in just a few years, or even do not use plastics.
Having said that, endorphins and lipoprotein trypsin peptide [βlph(61-69)] can reduce human addiction and tolerance to drugs.
You should know that after humans use many drugs for a long time, they will become dependent on drugs, that is, drug addictiveness, such as stimulants, analgesics, hallucinogens, etc.
Previously, scientists tried to reduce addiction through various means, such as cognitive behavioral therapy, oral topiramate treatment, oral dextamphetamine sustained release agents, etc.
But none of these methods have had a good effect.
This has a broader market than endorphins, and Zheng Li plans to wait and take it out later.
......
"You are here, I will take you to see the current general manager of the company. After that, I will stay in the laboratory downstairs for a long time. You may report to me about the company's business trends every week."
"You are usually responsible for following Cheng Gang's arrangements. Cheng Gang is our general manager. What you need to do is learn and become familiar with the company's operating model as soon as possible."
After Zheng Li took off his gloves and white coat, he came to his exclusive office upstairs and saw Li Miaomiao who had just landed on the ground and rushed over.
Li Miaomiao, who was originally nervous, calmed down after seeing Zheng Li, especially when he found that Zheng Lihe had not changed at all in school, he was still devoted to research.
She nodded and said, "Okay."
Zheng Li: "Cheng Gang, this is my classmate Li Miaomiao at Jiangcheng University. She mainly serves as my secretary. You usually call her if you have anything to do and take her to familiarize herself with the company's business conditions."
Cheng Gang looked at the very young girl in front of him and was pretty, and he thought to himself: Mr. Zheng is not trying to have an office romance, is he bringing his girlfriend to the company?
If this is true, Cheng Gang will lower his evaluation of Zheng Li at least two levels.
However, he was silent on the surface: "Okay, I will arrange Ms. Li. I am Cheng Gang, and I will work happily in the future."
Cheng Gang reached out to signal to shake hands with the other party. When Li Miaomiao shook hands, he said, "I would like to thank Mr. Cheng for his care in advance, and then I hope we can cooperate happily."
Zheng Li continued: "Cheng Gang, I know your questions, so I will make it clearer. Li Miaomiao and I are just classmates. To be precise, we are friends with good relationships, but not boyfriend and girlfriend."
